What exactly do you mean here? the question is very vague, and I read it in at least two ways.
I think @Olly means buying a second Avios ticket but adding to an existing Avios booking utilising the 2-4-1 voucher to get the second ticket for nothing (except taxes and fees)
The answer is no. You cannot apply a 2-4-1 to an existing booking.
The only way to do what they want is to use the 2-4-1 to buy a new pair of tickets (assuming reward seat availability) and then cancel the existing Avios booking for a refund of Avios and cash paid minus the £35 cancellation fee

If no-one knows the answer, the simplest thing to do would be to phone and ask them. Virgin are sometimes a bit more flexible than BA, but unless someone has actually done it, or tried to do it, we’re not going to know.
