Can one person stay behind for return Amex 241 flight?

We are flying to Japan in May with the 241 voucher. I am wondering if there is any possibility of one person staying behind for 2 more weeks on the return journey? If that’s not possible, can 1 person give up the return flights but the other fly back at planned?

Leaving one person behind is fine, as long and they’re not the voucher holder. The voucher holder needs to fly on the booking. However, I’m not sure how strictly that is enforced, especially at an outstation.
It’s fine, I’ve done it with my wife before when I (voucher holder) had to stay longer in KL & LA. No questions were asked of my wife at check in.
It’s not like skipping last leg of an ex EU flight where you’ve deprived BA of revenue.

Yes, BA won’t rebook you or refund their hefty surcharges hence they don’t care!
Just remember that, even if I doubt they’d check and Thegasman’s experience corroborates it, you’d be going against the T&Cs, therefore there is some risk involved, even if tiny.
I’ve also done this before, flying back from Japan in early 2020 – I was the voucher holder, and we also had connecting LHR->GLA flights. I think they might’ve asked my wife at checkin if I was with her, but she just said I was running late or something and all was fine. I was worried the domestic leg might get cancelled, but even that was fine. (I booked & paid for a separate flight back a week or two later.)
As others say, it does seem to be against the rules, so not risk free. For obvious reasons I didn’t even try to get a refund of the taxes on the unused flights (not entirely sure how much would’ve been due anyway), so BA got to keep the cash and BA had a spare seat to use.
