If I sign up to Amex platinum and use my wife as a supplementary holder, if I cancel the card can my wife apply for the platinum and then use me as a supplementary holder?

yes, but would make more sense to refer your wife first before cancelling your card

And get her supplementary after the issue of your main card, rather than during the application process (unless this has changed).
