If you accept a paid for upgrade in advance of arrival, does that automatically rule out any chances of a complimentary upgrade at check in?

Not sure what you’re asking. If you mean a further upgrade on top, I don’t why not, except that it’s unlikely that would have been offering the initial upgrade if they thought the cabin was going to be overbooked.
You certainly aren’t going to get your money back and a free upgrade instead.
I think if you apply for an e-upgrade at time of booking you can be pretty sure they will charge you, whether or not you get a complimentary one.
I just had one where they say they upgraded me 2 levels and so took the cash as well!
I would say only go for the paid upgrade if you really want to be sure to get a certain room type or view.
